The 2V6.11 cell line was derived from the human embryonic kidney cell line 293 [HEK-293] in 2001. The 293 [HEK-293] cells were transfected with the plasmid pVgRXR carrying a Zeocin-resistant marker to obtain 293pVgRXR cells. Subsequently, they were transfected with the plasmid pEKORF6 containing the Ad5 E4ORF6 gene encoding E4 24K. pEKORF6 was derived from the plasmid pIndHydro containing the hygromycin resistance gene, and the vector contained SV40 viral sequences. The 2V6.11 cell line was initially isolated by cloning rings from a culture medium containing hygromycin to obtain monoclonal cells. The human adenovirus E4 34 KDa protein induced and expressed by 2V6.11 cells can be detected by Western blotting; 2V6.11 cells can be used as a tool for studying adenovirus E4 oncogenes.
